Cancer Cell Engraftment

Cancer cell engraftment is the establishment and sustained growth of transplanted cancer cells within a recipient tissue or organism, providing a way to study tumor biology in vivo. After introduction into a suitable microenvironment, cancer cells must survive, attach to local structures, adapt to host signals, and proliferate sufficiently to form a detectable tumor. Researchers use engraftment in xenograft and patient-derived tumor models to investigate tumor initiation, growth, invasion, and treatment response. Successful engraftment can preserve important features of a cancer and support preclinical testing, although outcomes depend on cell properties, implantation site, host conditions, and interactions with the surrounding tissue.

We describe a method to significantly enhance orthotopic engraftment of lung cancer cells into the murine lungs by pre-conditioning the airways with injury. This approach may also be applied to study stromal interactions within the lung microenvironment, metastatic dissemination, lung cancer co-morbidities, and to more efficiently generate patient derived xenografts.

Determining donor cell engraftment presents a challenge in mouse bone marrow transplant models that lack well-defined phenotypical markers. We described a methodology to quantify male donor cell engraftment in female transplant recipient mice. This method can be used in all mouse strains for the study of HSC functions.
