Cd3-positive T Lymphocytes

CD3-positive T lymphocytes are immune cells defined by expression of the CD3 protein complex associated with the T-cell receptor, making them central mediators of adaptive cellular immunity. When the receptor recognizes antigen presented by major histocompatibility complex molecules, CD3 transmits activation signals that regulate T-cell proliferation, differentiation, cytokine production, and target-cell killing. In cancer research, measuring CD3-positive cells in tumors and blood helps characterize immune infiltration, assess responses to immunotherapy, and distinguish immune-active from immune-suppressed tumor environments. Their abundance and functional state can also inform biomarker development and the design of treatments that strengthen antitumor T-cell activity.

We present in the current study a novel fluorescence-based assay using lymphocytes derived from a transgenic mouse. This assay is suitable for high-throughput screening (HTS) of small molecules endowed with the capacity of either inhibiting or promoting lymphocyte activation.

Isolation and Th17 Differentiation of Naïve CD4 T Lymphocytes

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.
