Cell Culture Incubation

Cell culture incubation is the controlled maintenance of living cells under laboratory conditions that support growth, survival, and reproducible experimentation. In a humidified incubator, temperature, carbon dioxide concentration, and oxygen availability are regulated to stabilize the culture medium’s pH and mimic key aspects of the cellular environment, while nutrient-rich media supply essential substrates. In cancer research, incubation enables the expansion of tumor-derived cell lines, primary cancer cells, and three-dimensional cultures for studying proliferation, migration, signaling, and drug responses. Consistent conditions improve experimental reliability and support disease modeling, therapeutic screening, and investigations of how tumors interact with their microenvironment.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. This equipment meets clinical good manufacturing practice (cGMP) and clinical good lab practice (cGLP) guidelines.

Laboratory soil warming experiments usually employ two or more constant temperatures in multiple chambers. By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.

Responding for food or drug-paired cues increases over the course of a period of abstinence and this may relate to an increased susceptibility to relapse behaviors. Here we detail a procedure for evaluating this "incubation of craving" in rats that have self-administered sucrose.
