Cell Line Xenograft

A cell line xenograft is an in vivo cancer model created by implanting cultured tumor cells into an immunocompromised animal, typically a mouse, to study tumor growth and treatment response. Because reduced immune activity allows human cancer cells to survive and form tumors, researchers can monitor how malignant cells proliferate, interact with surrounding tissues, and respond to candidate therapies. Cell line xenografts support preclinical drug testing, dose and efficacy assessments, and investigations of therapeutic resistance. Although they may not fully reproduce patient tumors or immune responses, these models provide a controlled bridge between cell-based experiments and more complex cancer studies.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.

Germ Cell Transplantation and Testis Tissue Xenografting in Mice

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

This manuscript details a method used to generate prostate cancer patient derived xenografts (PDXs) from circulating tumor cells (CTCs). The generation of PDX models from CTCs provides an alternative experimental model to study prostate cancer; the most commonly diagnosed tumor and a frequent cause of death from cancer in men.
