Colorectal Liver Metastasis

Colorectal liver metastasis is the development of secondary tumors in the liver after colorectal cancer cells leave the primary tumor and establish growth at a distant site. This condition is clinically important because the liver is a frequent site of spread, and metastatic disease strongly influences treatment planning and prognosis. Tumor cells can invade blood vessels, enter the portal circulation, and lodge in hepatic tissue, where interactions with liver cells, immune populations, and extracellular matrix support colonization and tumor expansion. Detection and characterization through imaging, biopsy, and molecular analysis guide decisions about systemic therapy, surgical resection, ablation, or combined treatment, while research into metastatic niches and treatment resistance seeks to improve disease control and patient outcomes.

The ability of metastatic clones to colonize distant sites depends on their proliferation capacity and/or their ability to survive in the host microenvironment without significant proliferation. Here, we present an animal model that allows quantitative visualization of both types of liver colonization by metastatic clones.

Portal vein injection of colorectal cancer (CRC) organoids generates stroma-rich liver metastasis. This mouse model of CRC hepatic metastasis represents a useful tool to study tumor-stroma interactions and develop novel stroma-directed therapeutics such as adeno-associated virus-mediated gene therapies.
