Contrast Agent Delivery

Contrast agent delivery is the controlled administration and distribution of substances that enhance differences between tissues during medical imaging, enabling tumors and surrounding structures to be visualized more clearly. After intravenous or localized administration, an agent circulates through the body, reaches tissue through the vasculature, and alters signals detected by modalities such as magnetic resonance imaging, computed tomography, or ultrasound; its distribution can reflect blood flow, vascular permeability, or molecular binding. In cancer research, delivery strategies support tumor detection, characterization, treatment planning, and response monitoring, while targeted formulations may improve imaging specificity and reduce exposure to healthy tissue.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.

(2015)In this video, a water-filled microinjection setup consisting of a syringe and microinjector tubing is used for precise drug delivery. A rat with a pre-implanted guide cannula undergoes targeted brain microinjection, enabling accurate administration of therapeutic agents in awake animals with minimal tissue damage.

As neuroscience inquiry becomes more sophisticated, investigation of brain structures and circuitry requires improved levels of accuracy and higher resolution. We have developed a method for the preparation and implantation of a chronic infusion system within the brain utilizing a borosilicate microcannula with a tip diameter of 50 microns.
