Droplet Stabilizing Oil

Droplet stabilizing oil is an immiscible liquid phase used to form and preserve uniform aqueous droplets in microfluidic systems, where it supports controlled compartmentalization of biological reactions. As droplets move through channels, the oil surrounds each aqueous compartment, while dissolved surfactants lower interfacial tension and reduce coalescence, evaporation, and material exchange between neighboring droplets. In cancer research, this stability enables high-throughput analysis of individual cells, secreted molecules, nucleic acids, or drug responses within isolated microreactors. By maintaining consistent droplet size and composition, stabilizing oil improves assay reproducibility and supports applications such as single-cell screening, digital PCR, and studies of tumor heterogeneity.

Plasmid Stability Analysis with Open-Source Droplet Microfluidics

An accessible, open-source microfluidic workflow is presented for the parallelized analysis of plasmid retention in bacteria. By employing fluorescence microscopy to quantify plasmid presence in single-cell microcolonies within gel microdroplets, this method provides a precise, accessible, and scalable alternative to traditional plate counting.

We illustrate the methods involved in screening and identification of the biosurfactant producing microbes. Methods for chromatographic characterization and chemical identification of the biosurfactants, determining the industrial applicability of the biosurfactant in enhancing residual oil recovery are also presented.

Reconstitution of Basic Mitotic Spindles in Spherical Emulsion Droplets

The assembly and positioning of the mitotic spindle depend on the combined forces generated by microtubule dynamics, motor proteins and cross-linkers. Here we present our recently developed methods in which the geometrical confinement of spherical emulsion droplets is used for the bottom-up reconstitution of basic mitotic spindles.
