L-band Epr Spectrometer

An L-band EPR spectrometer is an electron paramagnetic resonance instrument that detects chemical species containing unpaired electrons, using relatively low-frequency microwaves near 1 GHz. It applies a magnetic field while monitoring microwave absorption, producing spectra that reveal the concentration, electronic environment, and interactions of paramagnetic centers. In cancer research, L-band EPR supports noninvasive or minimally invasive assessment of tissue oxygenation, tumor hypoxia, oxidative stress, and redox status through endogenous signals or paramagnetic probes. Its lower operating frequency can improve measurements in larger, heterogeneous biological samples, helping researchers characterize tumor physiology and evaluate responses to treatment.

The goal of this protocol is to use electron paramagnetic resonance (EPR) monitored redox titrations to identify different cofactors of Saccharomyces cerevisiae Nar1. Redox titrations offer a very robust way to obtain midpoint potentials of different redox active cofactors in enzymes and proteins.

This article describes methods for site-directed spin labeling and reconstitution of pentameric ligand-gated channels for Electron Paramagnetic Resonance studies. This protocol can be adapted for any membrane protein. The reconstitution method described here can also be used for patch-clamp measurements of macroscopic and single-channel currents in a defined lipid system.
