Lps Activation

LPS activation is the cellular response to lipopolysaccharide (LPS), a bacterial outer-membrane component that triggers innate immune signaling and can influence cancer-associated inflammation. In the canonical pathway, LPS is recognized by CD14 and transferred to the TLR4–MD-2 receptor complex, promoting receptor dimerization and downstream activation of NF-κB and mitogen-activated protein kinases. These pathways induce inflammatory mediators, including cytokines and chemokines, that can alter immune-cell behavior and the tumor microenvironment. In cancer research, controlled LPS stimulation helps investigators model inflammation, examine immune–tumor interactions, evaluate signaling inhibitors, and assess how bacterial products may affect tumor progression or responses to therapy.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

This video demonstrates a method to detect NLRP3 inflammasome activation in macrophages using fluorescent reporter probes. The cells are treated with potassium ionophores and glycine to induce inflammasome assembly and caspase-1 activation. Fluorescent reporter probes visualize activated caspase-1, and cells are stained with a fluorescent dye for nuclei visualization.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
