Prolonged Rna Knockdown

Prolonged RNA knockdown is a gene-silencing approach that reduces target RNA levels over an extended period, allowing researchers to study sustained changes in gene function. It typically uses small interfering RNA or short hairpin RNA to guide the RNA-induced silencing complex to complementary messenger RNA, promoting its cleavage or blocking translation; sustained delivery or expression maintains the effect. In cancer research, prolonged RNA knockdown helps assess how oncogenes, tumor suppressors, and drug-resistance pathways influence cancer-cell growth, survival, invasion, and treatment response. This strategy supports target validation and the development of RNA-based therapeutic approaches.

MicroRNAs play crucial roles in the brain and are potential targets for modeling neuro-degeneration. However, perturbing miRNA levels is challenging due to the short length of miRNA and inaccessibility of the brain tissue. This video presents a method for antagomir design and brain specific delivery using a neuropeptide in mice.
