Tumor Stroma

Tumor stroma is the noncancerous tissue and cellular environment surrounding and supporting tumor cells, making it a central component of the tumor microenvironment. It includes extracellular matrix, cancer-associated fibroblasts, blood vessels, and immune cells that communicate with cancer cells through cytokines, growth factors, and physical remodeling; these interactions can promote angiogenesis, invasion, immune suppression, and resistance to treatment. In cancer research, analyzing tumor stroma helps explain how tumors grow and spread, identify prognostic biomarkers, and reveal therapeutic targets beyond the malignant cells themselves. Stromal features are also important for predicting treatment response and developing strategies that alter the tumor microenvironment.

A Novel Stromal Fibroblast-Modulated 3D Tumor Spheroid Model for Studying Tumor-Stroma Interaction and Drug Discovery

A novel three-dimensional spheroid model based on the heterotypic interaction of tumor cells and stromal fibroblasts is established. Here, we present coculture of tumor cells and stromal fibroblasts, time-lapse imaging, and confocal microscopy to visualize the formation of spheroids. This three-dimensional model offers a pertinent platform to study tumor-stroma interactions and test cancer therapeutics.

This protocol presents a 3D biomimetic model with accompanying fibrotic stromal compartment. Prepared with physiologically relevant hydrogels in ratios mimicking the bio-physical properties of the stromal extracellular matrix, an active mediator of cellular interactions, tumor growth and metastasis.
