Benign Malignant Nodules

Benign and malignant nodules are localized tissue masses classified by whether their cells remain noninvasive or show cancerous behavior, making the distinction essential for clinical risk assessment and treatment planning. Clinicians evaluate a nodule’s size, growth pattern, borders, and internal features with physical examination and imaging, then use cytology or biopsy to assess cellular atypia, invasion, and other histopathologic findings. Benign nodules may require monitoring or symptom-directed treatment, whereas malignant nodules can need surgery, systemic therapy, or additional staging. Accurate classification supports early cancer detection, avoids unnecessary intervention, and guides individualized patient management across clinical specialties.

Three dimensional culture of mammary epithelial cells on a reconstituted basement membrane is a useful method to recapitulate the in vivo architecture of the benign breast, and to differentiate the malignant phenotype from the benign breast phenotype. Importantly, this system can be applied to study invasive carcinomas in other tissues.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.

Transgenic lck:eGFP zebrafish express GFP highly in T lymphocytes, and have been used to study T cell development and acute lymphoblastic leukemia. This line can be used to study B cells, which express lck at lower levels. This protocol describes purification of malignant and non-malignant B cells from lck:eGFP zebrafish.

Cooperation between an activated oncogene like RASV12 and mutations in cell polarity genes like scribbled, result in tumor growth in Drosophila where tumor cells also display invasive behaviors.
