Malignant Strictures

Malignant strictures are abnormal narrowings of a hollow organ caused by cancer, often obstructing the esophagus, bile duct, colon, or other passages and impairing normal flow. They develop when tumor growth infiltrates and compresses the surrounding wall, while inflammation and fibrosis can further reduce the lumen. Clinicians evaluate these lesions with imaging, endoscopy, tissue sampling, and assessment of symptoms such as dysphagia, jaundice, abdominal pain, or altered bowel function. Accurate identification distinguishes malignant from benign strictures and guides treatment, which may include cancer-directed therapy, endoscopic dilation, stent placement, or surgical intervention to relieve obstruction and improve quality of life.

In the present protocol, a urethral stricture induction was developed in Wistar rats, followed by urethral reconstruction with a buccal mucosa graft. A retrograde urethrogram and laser Doppler assessment were performed, validating urethral reconstruction (after stricture formation) and graft placement.

Transgenic lck:eGFP zebrafish express GFP highly in T lymphocytes, and have been used to study T cell development and acute lymphoblastic leukemia. This line can be used to study B cells, which express lck at lower levels. This protocol describes purification of malignant and non-malignant B cells from lck:eGFP zebrafish.
