Cell Purity Analysis

Cell purity analysis is the assessment of how closely a cultured, isolated, or differentiated cell population matches its intended identity and how much contamination it contains, an essential step in developmental biology. It works by measuring cell-type-specific markers and comparing signals from the target population with markers of unwanted cells, using approaches such as immunostaining, flow cytometry, microscopy, or gene-expression assays. These measurements help researchers verify differentiation protocols, track lineage specification, identify mixed populations, and improve the reproducibility of studies involving stem cells, organoids, and developing tissues. Reliable purity estimates also support downstream molecular analyses and functional experiments.

Here we describe a robust method of determining immune cell identity and purity through epigenetic signatures detected using quantitative PCR (qPCR). DNA demethylation at a specific locus serves as a unique identifier for a particular cell type and allows for identification of CD8+, regulatory, or Th17 T cells.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Cell Cycle Analysis

2023

Cell cycle refers to the set of events through which a cell grows, replicates its genome, and ultimately divides into two daughter cells through the process of mitosis. Because the amount of DNA in a cell shows characteristic changes throughout the cycle, techniques known as cell cycle analysis can be used to separate a population of cells according to the different phases of cell cycle they are in, based on their varying DNA content. This video will cover the principles behind cell cycle...
