Dolphin Spermatozoa

Dolphin spermatozoa are specialized haploid male gametes that deliver paternal genetic material to the egg and initiate embryonic development. Their streamlined head contains the nucleus and acrosome, while the flagellum generates motility that supports movement through the female reproductive tract; biochemical capacitation and the acrosome reaction enable sperm–egg binding and penetration. In developmental biology, studying dolphin sperm structure, maturation, motility, and fertilization provides insight into mammalian reproduction and early developmental events. These analyses also support reproductive health assessment, assisted reproduction, population management, and conservation programs for dolphin species.

This protocol describes a detailed methodology for detecting H2O2 localization within Solea senegalensis spermatozoa using a sensitive fluorochrome DCFH-DA for ROS, a live mitochondria stain for mitochondria, and DAPI for nuclei visualization, respectively.
