Follicle Cell Organization

Follicle cell organization is the spatial arrangement and coordinated behavior of somatic cells surrounding a developing oocyte, a structure that supports folliculogenesis and reproductive development. Follicle cells establish distinct layers and communicate with the oocyte through direct cell contacts and paracrine signaling, regulating nutrient transfer, growth, differentiation, and maturation. Their organization changes as the follicle develops, creating a specialized microenvironment that influences oocyte quality and hormone production. Studying these cellular interactions helps explain ovarian development, fertility, and reproductive disorders, while informing research on assisted reproduction, ovarian tissue models, and developmental mechanisms.

Isolating Hair Follicle Stem Cells and Epidermal Keratinocytes from Dorsal Mouse Skin

An ideal model for studying adult stem cell biology is the mouse hair follicle. Here we present a protocol for isolating different populations of hair follicles stem cells and epidermal keratinocytes, employing enzymatic digestion of mouse dorsal skin followed by FACS analysis.

Such follicles develop in culture, which allows for genetic and/or pharmacologic manipulations to be coupled with in vitro development assays and live imaging. Additionally, follicles can be used for molecular studies, such as isolating mRNA and protein.

A highly promising technique to generate tissue constructs without using matrix is to culture cells in a simulated microgravity condition. Using a rotary culture system, we examined ovarian follicle growth and oocyte maturation in terms of follicle survival, morphology, growth, and oocyte function under the simulated microgravity condition.
