Magnetic Microbead Separation

Magnetic microbead separation is a cell-isolation technique that uses magnetic particles to enrich or remove specific cell populations from a mixed sample. Antibodies or other binding molecules attached to the beads recognize surface markers, allowing labeled cells to be retained in a magnetic field while unlabeled cells are washed away, or enabling depletion of unwanted populations. In developmental biology, this method helps researchers isolate stem cells, progenitors, and lineage-specific cells for culture, molecular analysis, and differentiation studies. Its speed, scalability, and compatibility with viable cells support investigations of cell fate, tissue formation, and developmental signaling.

Low-density neutrophils (LDN) increase significantly in several diseases. LDN are usually isolated through cell sorting. We present a practical method to obtain pure and viable LDN. After density-gradient centrifugation of peripheral blood, cells are incubated with magnetic microbeads, and then LDN are separated through magnetic columns.

Here, we present a protocol to induce ocular hypertension in the murine eye that results in the loss of retinal ganglion cells as observed in glaucoma. Magnetic microbeads are injected into the anterior chamber and attracted to the iridocorneal angle using a magnet to block the outflow of aqueous humour.
