Marrow Stromal Cells

Marrow stromal cells are supportive connective-tissue cells in the bone marrow that organize the local environment required for blood-cell formation and tissue maintenance. They regulate this niche through cell-cell interactions, extracellular matrix production, and secretion of signaling molecules that influence hematopoietic stem and progenitor cells, while some populations can differentiate into bone, cartilage, and fat lineages. In developmental biology, studying marrow stromal cells reveals how skeletal tissues and blood-forming compartments develop together. Their roles in tissue repair, stem-cell regulation, and pathological remodeling also make them important for regenerative medicine and research into blood and bone disorders.

This video demonstrates a method for obtaining Schwann cell-like cells from multipotent bone marrow stromal cells (BMSCs). Initially, the BMSCs undergo hypoxia treatment, followed by directed differentiation into neuronal progenitor cells. Subsequently, specific media and culture conditions are employed to induce the transformation of these progenitor cells into Schwann cell-like cells.

A strategy to quantitatively analyze histological data in the bone marrow is presented. Confocal microscopy of fluorescently labeled cells in tissue sections results in 2-dimensional images, which are automatically analyzed. Co-localization analyses of different cell types are compared to data from simulated images, giving quantitative information about cellular interactions.
