Human Hematopoietic Engraftment

Human hematopoietic engraftment is the establishment and sustained function of transplanted human hematopoietic stem and progenitor cells within a recipient’s blood-forming tissues, enabling regeneration of blood and immune cells. After transplantation, these cells home to supportive bone marrow niches, adhere to local microenvironments, and undergo self-renewal and differentiation into multiple hematopoietic lineages. In developmental biology, engraftment assays help evaluate stem cell potential, lineage development, and interactions between human cells and their tissue environment. The approach also supports research on blood disorders, immune development, transplantation, and therapies that restore or modify human hematopoiesis.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

The colony forming cell (CFC) assay is an in vitro assay in which hematopoietic progenitors form colonies in a semi-solid medium. A combination of colony morphology, cell morphology, and flow cytometry are used to assess the ability of the progenitors to proliferate and differentiate along the different hematopoietic lineages.
