Zebrafish Blood Development

Zebrafish blood development is the formation and maturation of blood and blood-forming tissues in the zebrafish embryo, a transparent vertebrate model that reveals hematopoiesis in real time. During embryogenesis, mesodermal cells generate primitive erythrocytes and myeloid cells, while definitive hematopoietic stem and progenitor cells arise in the aorta-gonad-mesonephros region, enter circulation, and seed the kidney marrow, the adult hematopoietic organ. Studying these stages links cell fate specification, vascular development, and blood-cell migration, while fluorescent reporters and live imaging help researchers investigate conserved mechanisms of hematopoiesis, model blood disorders, and evaluate candidate therapies.

The zebrafish (Danio rerio) has become a popular model for studying genetics and developmental biology. The transparency of these animals at early developmental stages permits the direct visualization of tissue morphogenesis at the cellular level. Furthermore, zebrafish are amenable to genetic manipulation, allowing researchers to determine the effect of gene expression on the development of a vertebrate with a high degree of genetic similarity to humans. This video provides a brief overview of...

This protocol provides step-by-step instruction on how to generate parabiotic zebrafish embryos of different genetic backgrounds. When combined with the unparalleled imaging capabilities of the zebrafish embryo, this method provides a uniquely powerful means to investigate cell-autonomous versus non-cell-autonomous functions for candidate genes of interest.
