Water-in-oil Microdroplets

Water-in-oil microdroplets are small aqueous compartments dispersed within a continuous oil phase, providing controlled environments for chemical, biological, and materials-processing tasks in engineering. They form when immiscible water and oil streams are sheared in a microfluidic device, while surfactants stabilize the resulting interfaces and help produce uniform droplets. Each droplet can isolate reagents, cells, or particles, enabling parallel reactions, concentration control, and reduced sample consumption. These properties support high-throughput screening, digital assays, encapsulation, and microscale materials synthesis, while offering precise control over transport, reaction conditions, and product size.

Here, we demonstrate a simple production method for size-controllable, monodisperse, water-in-oil (W/O) microdroplets using a capillary-based centrifugal microfluidic device. This method requires only a small sample volume and enables high-yield production. We expect this method will be useful for rapid biochemical and cellular analyses.

Source: Jian, X., et al., Automated Microbial Cultivation and Adaptive Evolution using Microbial Microdroplet Culture System (MMC). J. Vis. Exp. (2022).This video demonstrates the use of a microbial microdroplet culture system to drive adaptive evolution of bacteria toward methanol tolerance. By mixing engineered bacterial cultures with a high-methanol stress medium and segmenting them into microdroplets, the system selects for mutants that exhibit improved growth under methanol stress. This manuscript describes a simple and high-throughput method for assembling water-soluble proteins with hydrophobic pigments that is based on water-in-oil emulsions. We demonstrate the effectiveness of the method in the assembly of native chlorophylls with four variants of recombinant water-soluble-chlorophyll binding proteins (WSCPs) of Brassica plants expressed in E. coli.
