Fibroblast Fusion

Fibroblast fusion is the joining of two or more fibroblasts to form a single multinucleated cell, providing a way to study cell identity, nuclear interactions, and genome function. Fusion can be induced experimentally with agents such as polyethylene glycol or by physiological fusogenic mechanisms that bring plasma membranes together, merge their lipid bilayers, and mix cytoplasmic contents; the nuclei may remain separate initially before combining. In genetics, fibroblast heterokaryons and hybrid cells help reveal complementation between mutant genomes, chromosome behavior, gene regulation, and factors that control cellular reprogramming. These models connect cell biology with disease mechanisms and regenerative research.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.
