Single-gene Analysis

Single-gene analysis is the focused investigation of one gene to determine its sequence, function, expression, or contribution to a genetic trait or disorder. Depending on the research question, it may compare DNA sequences, identify variants, measure gene expression, or trace inheritance patterns, linking changes in the gene to molecular and phenotypic effects. In genetics, this approach supports the study of gene function, mutation effects, and genotype–phenotype relationships. It can also aid variant interpretation, diagnosis of monogenic conditions, and the design of targeted experiments, providing detailed insight that broader genomic surveys may not capture.

Fluorescent in situ hybridization (FISH) to identify mRNA transcripts in individual cells allows analysis of polygenic activity such as the simultaneous transcription of more than one member of the var multigene family in Plasmodium falciparum infected erythrocytes 1. The technique is adaptable and can be used on different types of genes, cells and organisms.

We describe how to use laser capture microdissection (LCM) to obtain enriched populations of hippocampal neurons or single neurons from frozen sections of the injured rat brain for subsequent gene expression analysis using quantitative real time PCR and/or whole-genome microarrays.

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.
