Asc Focus Formation

ASC focus formation is the assembly of apoptosis-associated speck-like protein containing a CARD (ASC) into distinct intracellular puncta, providing a visible indicator of inflammasome activation. Following detection of infection- or danger-associated signals, sensor proteins recruit ASC through pyrin domain interactions, while ASC’s CARD domains promote clustering and recruitment of pro-caspase-1; activated caspase-1 then processes inflammatory cytokines and can trigger pyroptosis. Imaging and quantifying ASC foci helps researchers assess inflammasome activity in infected or stimulated cells, compare immune responses, and investigate how innate immune signaling contributes to inflammation, host defense, and infectious disease.

An In Vitro Technique to Study Inflammasome Complex Formation in Macrophages

This video demonstrates a technique to study NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome formation in primary macrophages via immunofluorescence. The macrophages are first primed with bacterial lipopolysaccharide (LPS) to upregulate NLRP3 protein expression. Exposing the macrophages to nigericin—a potassium ionophore—leads to the activation of NLRP3 and the formation of inflammasomes, which are visualized via immunofluorescence.
