Automated Colony Counter

An automated colony counter is a laboratory instrument that uses imaging and software to detect and enumerate microbial colonies growing on agar, improving the speed and consistency of viable-cell measurements. The system captures an image of a culture plate, distinguishes colonies from the background using features such as contrast, size, shape, and color, and applies preset or user-adjusted analysis parameters to calculate the count. In immunology and infection research, automated counting supports bacterial and fungal quantification, antimicrobial susceptibility studies, pathogen growth monitoring, and plaque or viability assays. Standardized analysis can reduce observer variability and improve reproducibility across experiments.

The Moxi Z miniature automated cell counter is a novel instrument that combines the Coulter Principle with patented thin-film sensor technology and a proprietary software algorithm to perform sizing and counting of a broad size range of particles as well as to determine the overall health of monodisperse mammalian cell cultures. In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
