Cd34+ Stem Cells

CD34+ stem cells are hematopoietic stem and progenitor cells identified by expression of the CD34 surface marker, making them important for studying blood formation and immune-system development. They reside primarily in bone marrow and can self-renew while producing progenitors that differentiate into lymphoid and myeloid cells, including leukocytes involved in host defense. Researchers isolate and enrich these cells using antibody-based magnetic separation or flow cytometry to analyze their function, infection responses, and capacity for immune reconstitution. In immunology and infection research, CD34+ cells support studies of hematopoiesis, pathogen effects on blood-cell development, transplantation, and strategies for restoring immunity after disease or treatment.

This study has established a stable and efficient method for the isolation, culture, and functional determination of vascular wall-resident CD34+ stem cells (CD34+ VW-SCs). This easy-to-follow and time-effective isolation method can be utilized by other investigators to study the potential mechanisms involved in cardiovascular diseases.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

The applications of this protocol include investigations on the effect of myeloid disease mutations or small molecules on myeloid differentiation of the CD34+ cells.
