Cell Fixation Permeabilization

Cell fixation and permeabilization is a sample-preparation technique that preserves cellular structure while allowing probes to access targets inside cells, making it essential for immunological and infection research. Fixatives stabilize or cross-link cellular components, and subsequent treatment with detergents or alcohols disrupts lipid membranes so antibodies and other labeling reagents can enter the cytoplasm or nucleus. Researchers use this approach to detect intracellular cytokines, pathogen proteins, nucleic acids, and signaling molecules by fluorescence microscopy or flow cytometry. Careful control of fixative type, exposure time, and permeabilization conditions helps maintain morphology and antigen accessibility, improving the accuracy of cellular phenotyping and host-pathogen studies.
