Cell Phenotyping

Cell phenotyping is the systematic characterization of cells by their physical features, molecular markers, functions, and responses to environmental signals. In immunology and infection research, techniques such as flow cytometry, microscopy, and single-cell analysis measure surface or intracellular markers, cytokine production, viability, and changes in cell state to distinguish immune cell subsets and pathogen-infected cells. These measurements help researchers track immune activation, differentiation, exhaustion, and host-pathogen interactions across tissues and disease stages. Cell phenotyping supports infection diagnosis, vaccine evaluation, immunotherapy development, and investigation of how pathogens alter cellular behavior, providing a detailed view of immune responses at the cellular level.

Primary cilia influence various signaling pathways. The mammalian cochlea is ideal for examining planar cell polarity (PCP) signaling. Cilia dysfunction affects cochlear outgrowth, cellular patterning and hair cell orientation, readouts of PCP. Our goal is to analyze PCP signaling in mouse cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy.

A Non-invasive Way to Isolate and Phenotype Cells from the Conjunctiva

The exposed normal ocular surface consists of cornea and conjunctiva. Epithelial cells, goblet cells and immune cells are present in the conjunctiva. Here, a non-invasive, technique of impression cytology is described using an impression cytology device and flow cytometry to analyze immune cells in the conjunctiva.

Preparation of a Micropatterned Substrate to Study Schwann Cell Phenotypes

The video describes the preparation of a micropatterned substrate to study cell phenotype specification. A protein-coated micropatterned PDMS stamp is used to transfer the micropattern on a PDMS-coated coverslip. The coverslip is then treated with a surfactant to hinder cell adhesion at the unwanted surfaces. Cells are then added to settle in the pattern and incubated for further analysis.
