Nasal Extract Analysis

Nasal extract analysis is a laboratory method for characterizing the molecular and cellular contents of nasal secretions, providing a noninvasive window into local airway immunity and infection. The process typically involves collecting nasal material, extracting soluble components, and measuring targets such as antibodies, cytokines, antimicrobial factors, nucleic acids, or pathogen-associated signals with biochemical or immunological assays. In immunology and infection research, these measurements help assess mucosal immune responses, identify infection-related changes, evaluate biomarkers, and monitor responses to vaccines or treatments. Because sampling reflects activity at the respiratory surface, the method can complement blood-based analyses and support studies of host-pathogen interactions.

2025

Proteinases are vital for bacterial pathogenesis, enabling invasion and immune evasion. Single-step extraction of cytoplasmic and membrane proteins of bacteria in their active form is harrowing. This video article demonstrates the extraction of whole cellular proteins of Leptospira for zymography analysis to detect and characterize gelatinases.

Culturing of Human Nasal Epithelial Cells at the Air Liquid Interface

Nasal epithelial cells, obtained through superficial scrape biopsy of human volunteers, are expanded and transferred onto tissue culture inserts. Upon reaching confluency, cells are grown at air liquid interface, yielding cultures of ciliated and non-ciliated cells. Differentiated nasal epithelial cell cultures provide viable experimental models for studying the respiratory mucosa.
