Cuticle Peeling Technique

The cuticle peeling technique is a sample-preparation method that removes the tough external cuticle of invertebrate hosts to expose underlying tissues, enabling direct analysis of host defense and pathogen interactions. Controlled mechanical or chemical disruption separates the cuticular layer while preserving accessible internal structures for microscopy, staining, or molecular assays. In immunology and infection research, this approach can improve visualization of immune cells, microbial localization, tissue damage, and barrier responses that are obscured by the intact surface. By linking external barrier integrity with internal infection phenotypes, cuticle peeling supports studies of innate immunity, host susceptibility, and pathogen invasion.

Aerial plant organs are protected by the cuticle, a supramolecular biopolyester-wax assembly. We present protocols to monitor selective removal of epi- and intracuticular waxes from tomato fruit cuticles on molecular and micro scales by solid-state NMR and atomic force microscopy, respectively, and to assess the cross-linking capacity of engineered cuticular biopolyesters.

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.
