Cytokine Concentration Analysis

Cytokine concentration analysis is the quantitative measurement of signaling proteins released by immune and other cells, providing insight into inflammation, immune activation, and infection. Common methods use antibody-based assays, such as ELISA or multiplex bead-based platforms, in which capture and detection antibodies bind a target cytokine and generate a signal compared with a calibrated standard curve. In immunology and infection research, measuring cytokines in serum, plasma, cell-culture supernatants, or tissue samples helps characterize host responses, compare disease states, evaluate treatments, and identify patterns of immune dysregulation. Accurate sample handling and assay controls are essential because cytokine levels can vary with timing, processing, and biological context.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.

We illustrate the application of 1H(15N,αγ)12C resonant nuclear reaction analysis (NRA) to quantitatively evaluate the density of hydrogen atoms on the surface, in the volume, and at an interfacial layer of solid materials.
