Dual Channel Imaging

Dual channel imaging is a microscopy approach that records two distinct optical signals from the same specimen, allowing researchers to compare cellular structures or biological events in spatial and temporal context. In practice, separate fluorescent labels emit at different wavelengths, and filters or detectors isolate each channel for synchronized visualization and analysis. In immunology and infection research, dual channel imaging can distinguish host immune cells from microbial targets, track their interactions, and assess co-localization, recruitment, or changes in protein or organelle distribution. These measurements help connect molecular localization with immune function, pathogen behavior, and treatment responses.

2013

Dual camera emission splitting systems for two-color fluorescence microscopy generate real-time image sequences with exceptional optical and temporal resolution, a requirement of certain live cell assays including parallel plate flow chamber adhesion assays. When software is employed to merge images from simultaneously acquired emission channels, pseudocolored image sequences are produced.

Source: Schott, J. T., et al. Preparation of Rat Oligodendrocyte Progenitor Cultures and Quantification of Oligodendrogenesis Using Dual-infrared Fluorescence Scanning. J. Vis. Exp. (2016). This video demonstrates the use of dual fluorescence imaging to evaluate the differentiation of oligodendrocytes from precursor cells (OPCs) isolated from a rat pup brain. The process includes fixing the cells, labeling specific markers with primary and fluorophore-conjugated secondary antibodies, and...
