Ethanol Resistance

Ethanol resistance refers to the ability of microorganisms to survive or tolerate exposure to ethanol, a property important for interpreting alcohol disinfection and infection-control practices. Ethanol disrupts lipid membranes, denatures proteins, and damages cellular structures; resistant or tolerant organisms may reduce ethanol penetration, alter membrane composition, activate stress responses, or persist in protective communities, although responses vary with species and exposure conditions. In immunology and infection research, measuring survival across ethanol concentrations and contact times helps distinguish true resistance from inadequate exposure and evaluate disinfectant performance. This knowledge supports hygiene protocols, laboratory biosafety, and studies of microbial persistence in clinical and environmental settings.

Ethanol Resistance Assay to Evaluate Biofilm Matrix Integrity

Source: Bucher, T., et al. Methodologies for Studying B. subtilis Biofilms as a Model for Characterizing Small Molecule Biofilm Inhibitors. J. Vis. Exp. (2016).This video demonstrates how biofilm matrix integrity influences bacterial resistance to ethanol by comparing colony-forming units in untreated and inhibitor-disrupted biofilms. Reduced CFUs in inhibitor-treated samples demonstrate that the extracellular matrix is essential for protecting biofilm-embedded cells from antimicrobial stress.

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Biofuels: Producing Ethanol from Cellulosic Material

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University In this experiment, cellulosic material (such as corn stalks, leaves, grasses, etc.) will be used as a feedstock for the production of ethanol. The cellulosic material is first pretreated (ground and heated), digested with enzymes, and then fermented with yeast. Ethanol production is monitored using an ethanol probe. The experiment can be extended to optimize ethanol production by varying the feedstock used,...

An Assay for Measuring the Effects of Ethanol on the Locomotion Speed of Caenorhabditis elegans

C. elegans is a useful model for studying the effects of ethanol on behavior. We present a behavioral assay that quantifies the effects of ethanol on the locomotion speed of crawling worms; both initial sensitivity and the development of acute functional tolerance to ethanol can be measured with this assay.
