Hemagglutinin Stalk Targeting

Hemagglutinin stalk targeting is an influenza research strategy that focuses immune responses or antiviral agents on the conserved stalk region of hemagglutinin, a viral surface protein required for host-cell entry. After influenza virions enter endosomes, acidic conditions trigger hemagglutinin to undergo a structural rearrangement that drives fusion between viral and cellular membranes; stalk-binding antibodies can interfere with this transition and prevent infection. Because the stalk is generally more conserved than the variable head, targeting it may support broadly protective vaccines and antibody-based therapies across multiple influenza strains. This approach informs universal influenza vaccine design and the study of viral immune evasion.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

A simple slot blot method was developed for the quantification of influenza viral hemagglutinin and neuraminidase using universal antibodies targeting their most conserved sequences identified through bioinformatics analyses. This innovative approach may provide a useful alternative to quantitative determination of all viral hemagglutinin and neuraminidase.
