In Vivo Transfer

In vivo transfer is a research method that introduces cells, immune components, or infectious material into a living organism to examine their behavior and effects in a physiological setting. In immunology, investigators commonly transfer defined donor immune cells into recipient animals, where the cells can circulate, migrate to tissues, expand, and respond to antigen or infection. This approach helps distinguish cell-intrinsic immune functions from signals provided by the surrounding host environment. In vivo transfer supports studies of lymphocyte development, immune memory, host-pathogen interactions, and disease mechanisms, while enabling direct assessment of immune protection, pathology, and therapeutic potential.

Gene Transfer to the Developing Mouse Inner Ear by In Vivo Electroporation

The mouse inner ear is a placode-derived sensory organ whose developmental program is elaborated during gestation. We define an in utero gene transfer technique consisting of three steps: mouse ventral laparotomy, transuterine microinjection, and in vivo electroporation. We use digital video microscopy to demonstrate the critical experimental embryological techniques.

Here, we present an in vivo technique for gene transfer to Schwann cells (SCs) in the rodent sciatic nerve. This simple technique is useful for investigating signaling mechanisms involved in the development and maintenance of myelinating SCs.

This method is to introduce a transgene into the endothelium of rabbit carotid arteries. Introduction of the transgene allows the assessment of the biological role of the transgene product either in normal arteries or disease models. The method is also useful for measuring activity of DNA regulatory sequences.

Ex vivo ERG can be used to record electrical activity of retinal cells directly from isolated intact retinas of animals or humans. We demonstrate here how common in vivo ERG systems can be adapted for ex vivo ERG recordings in order to dissect the electrical activity of retinal cells.

We describe a general protocol for in vivo cell tracking using MRI in a mouse model with ex vivo labeled cells. A typical protocol for cell labeling, image acquisition processing and quantification is included.
