Inflammation Imaging

Inflammation imaging is the visualization of immune activation and tissue changes that occur during injury, infection, or disease, enabling researchers and clinicians to assess inflammation in living systems. Imaging methods detect features such as increased blood flow, vascular permeability, immune-cell recruitment, or molecular signals using optical, ultrasound, magnetic resonance, computed tomography, or nuclear techniques, often with targeted contrast agents or probes. In immunology and infection research, these approaches reveal where and when inflammatory responses begin, how they spread, and how they resolve. The resulting spatial and temporal information supports disease characterization, therapeutic evaluation, and the development of more precise anti-inflammatory and antimicrobial strategies.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.

Overview In response to tissue injury and infection, mast cells initiate inflammation. Mast cells release chemicals that increase the permeability of adjacent blood capillaries and attract additional immune cells to the wound or site of infection. Neutrophils are phagocytic leukocytes that exit the bloodstream and engulf invading microbes. Blood clotting platelets seal the wound and fibers create a scaffold for wound healing. Macrophages engulf aging neutrophils to end the acute inflammatory...

Vascular calcification is an important predictor of and contributor to human cardiovascular disease. This protocol describes methods for inducing calcification of cultured primary vascular smooth muscle cells and for quantifying calcification and macrophage burden in animal aortas using near-infrared fluorescence imaging.
