Lipidomic Analysis

Lipidomic analysis is the comprehensive study of lipids in biological samples, including their identities, abundances, and structural changes, to reveal how lipid metabolism influences health and disease. It typically combines lipid extraction with chromatographic separation and mass spectrometry, which distinguishes lipid species by their chemical properties and mass-to-charge ratios for qualitative and quantitative profiling. In immunology and infection research, this approach tracks changes in membrane lipids, inflammatory mediators, and pathogen-derived lipids during immune activation or infection. These profiles can clarify host–pathogen interactions, identify biomarkers, and reveal metabolic pathways that may guide therapeutic development.

This protocol describes a unique clinical protocol for collecting human tear fluid samples using phenol red threads and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based workflow to discover the tear lipidomic profile. The simple methyl tert-butyl ether (MTBE)/methanol biphasic separation method enables rapid tear lipid extraction with high recovery for tear biomarker discovery.
