Lymphocyte Stimulation

Lymphocyte stimulation is the activation of B cells and T cells by signals that initiate adaptive immune responses to pathogens and other antigens. Antigen recognition through B-cell or T-cell receptors, together with co-stimulatory interactions and cytokine signaling, promotes lymphocyte proliferation, differentiation, and effector function; the exact response depends on the cell type and surrounding immune conditions. In immunology and infection research, stimulation assays help measure immune cell responsiveness, identify defects in host defense, and evaluate how vaccines, pathogens, or therapeutic agents influence antibody production and cellular immunity. These studies clarify mechanisms of immune protection and dysregulation.

An In Vitro Technique to Stimulate Lymphocytes via Pathogenic Bacteria

This video demonstrates an in vitro technique to stimulate lymphocytes via non-typeable Haemophilus influenzae (NTHi) and to assess the immune response. Incubating isolated peripheral blood mononuclear cells (PBMCs) with NTHi causes the bacterial cells to stimulate T lymphocytes, leading to cytokine production. Upon inhibiting the secretion of cytokines, immunofluorescence staining is performed to detect the accumulated intracellular cytokines in stimulated T lymphocytes.

Lymphocytes are the major players in adaptive immune responses. Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.
