Monocyte Recruitment

Monocyte recruitment is the directed movement of circulating monocytes from the bloodstream into tissues, where these innate immune cells help coordinate inflammation and host defense. During infection or tissue injury, inflammatory signals such as chemokines activate vascular endothelium, promote monocyte adhesion through integrins, and guide cells across the vessel wall by chemotaxis. Once recruited, monocytes can differentiate into macrophages or inflammatory dendritic cells, phagocytose pathogens and debris, and produce cytokines that shape local immune responses. Studying this process helps explain protective immunity, chronic inflammation, and infection-associated tissue damage, while informing research on therapies that modulate leukocyte trafficking.

Simultaneous Study of the Recruitment of Monocyte Subpopulations Under Flow In Vitro

Here, we present an integrated protocol that measures monocyte subpopulation trafficking under flow in vitro by use of specific surface markers and confocal fluorescence microscopy. This protocol can be used to explore sequential recruitment steps as well as to profile other leukocyte subtypes using other specific surface markers.

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.
