Multiplex Pcr

Multiplex PCR is a molecular technique that amplifies multiple DNA targets in a single reaction, making it useful for detecting several genetic markers efficiently. It uses multiple primer pairs under shared thermal-cycling conditions, with each pair selectively binding and copying a specific target sequence; the resulting amplicons can be distinguished by size, probe, or fluorescence. In immunology and infection research, multiplex PCR supports simultaneous identification of pathogens, detection of coinfections, and analysis of immune-response or antimicrobial-resistance markers. By conserving sample, reagents, and processing time, it enables broader diagnostic screening and more comprehensive investigation of infectious disease.

We demonstrate a simple multiplex PCR assay for quick-screening and typing of Staphylococcal Cassette Chromosome mec (SCCmec) types I-V for methicillin-resistant Staphylococcus aureus, and provide some of the vital steps and procedural nuances that make it successful for adapting this assay to individual laboratories.

Here we present a protocol for the measurement of relative telomere length (TL) using the monochrome multiplex quantitative polymerase chain reaction (MMqPCR) assay. The MMqPCR assay is a repeatable, efficient, and cost-effective method for measuring TL from human DNA in population-based studies.
