Neonatal Immune System

The neonatal immune system is the network of innate and adaptive defenses present during the first weeks of life, when newborns must resist infection while adapting to microbes outside the womb. Innate barriers, phagocytes, complement, and maternal antibodies provide early protection, whereas neonatal antigen-presenting cells and lymphocytes develop immune memory more gradually; exposure to antigens, cytokines, and microbial signals helps shape this transition. Understanding these age-dependent responses clarifies why newborns face distinctive risks from infection and why vaccination schedules, maternal immunization, diagnostic testing, and infection-control strategies are tailored to early life.

A technique to isolate cholangiocytes from the extrahepatic bile ducts of neonatal mice is described. The ducts are meticulously dissected, and then cells are isolated by outgrowth in thick collagen gels. This method provides a useful tool for studying extrahepatic bile duct development and pathology.

Modeling Infection in Neonatal Mice Using Encapsulated Bioluminescent Bacteria

Source: Seman, B. G., et al. A Neonatal Imaging Model of Gram-Negative Bacterial Sepsis. J. Vis. Exp. (2020)The video demonstrates a neonatal mouse model for studying systemic infection using encapsulated bioluminescent bacteria. Neonatal pups are injected with high- or low-dose bacterial suspensions and returned to their mother for postnatal care. The encapsulated bacteria evade early immune detection, spread systemically, and are visualized over time using luminescence imaging. This model...

Presented here is a protocol to perform comprehensive neonatal echocardiography by trained neonatologists in the neonatal intensive care unit. The trained individuals provide longitudinal assessments of heart function, systemic and pulmonary hemodynamics in a consultative role. The manuscript also describes the requirements to become a fully trained neonatal hemodynamics specialist.

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.
