Ph-sensitive Reporter

A pH-sensitive reporter is a fluorescent or otherwise detectable probe whose signal changes with acidity, enabling researchers to measure pH in cells, organelles, or other biological environments. Its mechanism relies on pH-dependent protonation of the reporter, which alters chromophore ionization and changes fluorescence intensity or spectral properties. In immunology and infection research, these reporters help track acidification in endosomes and phagosomes, monitor pathogen entry and intracellular trafficking, and assess how immune cells modify their microenvironment. Such measurements connect local pH dynamics with antimicrobial activity, pathogen survival, and cellular responses, providing a noninvasive way to study host-pathogen interactions over time.

Mitophagy, the selective degradation of mitochondria, has been implicated in mitochondrial homeostasis and is deregulated in various human diseases. However, convenient experimental methods for measuring mitophagy activity are very limited. Here, we provide a sensitive assay for measuring mitophagy activity using flow cytometry.
