Pulmonary Granuloma Formation

Pulmonary granuloma formation is an organized immune response in lung tissue that walls off persistent pathogens, foreign material, or other difficult-to-clear stimuli. Antigen-presenting cells activate T lymphocytes, whose cytokines recruit and sustain macrophages; these cells cluster, transform into epithelioid macrophages and multinucleated giant cells, and may become surrounded by lymphocytes and fibrotic tissue. In infections such as tuberculosis, this structure can restrict microbial spread, although impaired immunity or excessive inflammation may allow progression, tissue damage, or caseous necrosis. Studying pulmonary granulomas helps explain host-pathogen interactions, interpret lung pathology, and guide research into diagnostic markers and therapies for chronic infectious and inflammatory diseases.

In order to understand the cellular and molecular mechanisms underlying neotissue formation and stenosis development in tissue engineered heart valves, a murine model of heterotopic heart valve transplantation was developed. A pulmonary heart valve was transplanted to recipient using the heterotopic heart transplantation technique.

Selective Harvesting of Marginating-pulmonary Leukocytes

Marginating-pulmonary leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe selective harvesting of this subpopulation of pulmonary leukocytes, by forced perfusion of the lungs in rats and mice. Marginating-pulmonary leukocytes seem critical in determining susceptibility to blood-borne and lung-related diseases.

Hypoxic pulmonary vasoconstriction (HPV) is an important physiological phenomenon by which at alveolar hypoxia lung perfusion is matched to ventilation. The major vascular segment contributing to HPV is the intra-acinar artery. Here, we describe our protocol for the analysis of HPV of murine pulmonary vessels with diameters of 20-100 μm.
