Rna Toxicity Screening

RNA toxicity screening is the systematic evaluation of harmful cellular or immune responses caused by RNA molecules, an essential step in developing safe RNA-based therapeutics and vaccines. In immunology and infection research, screening typically exposes cultured cells or immune models to candidate RNA and measures outcomes such as cell viability, inflammatory signaling, cytokine release, and innate immune activation through pattern-recognition receptors. These assays can identify sequence-, structure-, dose-, or delivery-dependent toxicity and help distinguish intended immune stimulation from damaging responses. The results guide RNA design, chemical modification, formulation, and dosing while supporting the development of safer treatments, vaccine platforms, and infection-focused research tools.

A highly sensitive ribozyme-based assay, applicable to high-throughput screening of chemicals targeting the unique process of RNA editing in trypanosomatid pathogens, is described in this paper. Inhibitors can be used as tools for hypothesis-driven analysis of the RNA editing process and ultimately as therapeutics.

In vitro assays to measure virus replication have been greatly improved by the development of recombinant RNA viruses expressing luciferase or other enzymes capable of bioluminescence. Here we detail a high-throughput screening pipeline that combines such recombinant strains of measles and chikungunya viruses to isolate broad-spectrum antivirals from chemical libraries.
