Tear Volume Measurement

Tear volume measurement is the quantitative assessment of the amount of tear fluid produced or present on the ocular surface, supporting evaluation of lacrimal function and eye health. In the Schirmer test, a calibrated absorbent strip is placed along the lower eyelid for a defined period, and the length moistened by tears provides an estimate of aqueous tear production; capillary collection offers another measurement approach. In immunology and infection research, tear volume assessment helps characterize dry-eye conditions, standardize tear-fluid sampling, and support analysis of antimicrobial proteins, antibodies, inflammatory mediators, and other host-defense factors.

This video demonstrates the phenol red thread test to determine tear volume in an autoimmune dry eye rat model. The pH-sensitive acidic phenol red thread changes color when it comes in contact with the alkaline tears. The shorter length of color change in the phenol red thread corresponds to reduced tear volume.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.
