Therapeutic Antibody Production

Therapeutic antibody production is the development and manufacture of antibodies designed to prevent, diagnose, or treat disease, including infections and immune disorders. It typically begins by selecting an antibody that recognizes a target antigen, then expressing its encoding genes in cultured mammalian cells, where the cells assemble and secrete the antibody for purification and quality testing. Production must preserve antigen-binding specificity, molecular stability, and consistent activity while controlling impurities and unwanted immune responses. In immunology and infection research, these antibodies can neutralize pathogens or toxins, block host-pathogen interactions, guide immune effector functions, and support targeted therapies and diagnostics.

This protocol describes the production of a therapeutic antibody in a mammalian expression system. The methods described include preparation of vector DNA, stable transfection and serum-free adaptation of a human embryonic kidney 293 cell line, set up of large scale cultures and purification using affinity chromatography.

Antibody Production from a Batch Overgrowth Culture of Transfected Mammalian Cells

The video demonstrates a method for producing therapeutic antibodies using stably transfected mammalian cells. Tryptone supplementation stimulates cell division, resulting in overgrowth and supporting a higher antibody yield. The antibody-containing medium is harvested, filtered, and stored for further use.

This protocol describes the in vitro comparison of two key functional characteristics of rituximab: target binding and complement-dependent cytotoxicity (CDC) induction. The methods were employed for a side-to-side comparison between reference rituximab and a rituximab biosimilar. These assays can be employed during biosimilar development or as a quality control in their production.
