Viral Dna Extraction

Viral DNA extraction is a laboratory method that isolates DNA from virus-containing samples for detection, characterization, and downstream analysis. The process typically disrupts viral particles, releases their genomes, and separates DNA from proteins, lipids, cellular debris, and enzymatic inhibitors through chemical lysis, binding to a purification matrix, washing, and elution. In immunology and infection research, the resulting DNA supports PCR-based detection, viral load measurement, sequencing, and strain identification. Reliable extraction preserves genome integrity and removes contaminants that could interfere with amplification, helping researchers investigate transmission, monitor infections, evaluate diagnostic assays, and study host-pathogen interactions.

We describe the process of isolating high purity herpesvirus nucleocapsid DNA from infected cells. The final DNA captured from solution is of high concentration and purity, making it ideally suited for high-throughput sequencing, high fidelity PCR reactions, and transfections to produce new viral recombinants.

We describe an efficient method to separate single-stranded DNA, double-stranded DNA and RNA molecules from environmental viral communities. Nucleic acids are fractionated using hydroxyapatite chromatography with increasing concentrations of phosphate-containing buffers. This method permits the isolation of all viral nucleic acid types from environmental samples.
