Abdominal Aortic Constriction

Abdominal aortic constriction is a surgical method that partially narrows the abdominal aorta to create controlled pressure overload in the cardiovascular system. The reduced vessel diameter increases resistance to blood flow and raises left ventricular afterload, prompting compensatory cardiac hypertrophy and, with sustained stress, ventricular remodeling and dysfunction. In medicine and biomedical research, this procedure is commonly used in animal models to study the progression from pressure overload to heart failure and to investigate molecular mechanisms of cardiac adaptation. It also provides a reproducible platform for evaluating potential therapies that target hypertrophy, fibrosis, vascular regulation, or cardiac remodeling.

Transverse aortic constriction (TAC) in the mouse is a commonly used experimental model to study mechanisms underlying cardiac hypertrophy and heart failure development. Here, we describe procedures to constrict the aorta to create a reproducible degree of cardiac hypertrophy in mice.

Minimally invasive transverse aortic constriction (MTAC) conserves the essentials of regular transverse aortic constriction (TAC) while eliminating the use of a ventilator with tracheal intubation. It proves to be a highly desirable method for high-throughput studies on left ventricular overload, particularly in translational studies.

This video shows how to induce abdominal aortic aneurysms (AAA) in mice via transient intraluminal infusion of porcine pancreatic elastase into the infrarenal segment of the abdominal aorta. The model has the ability to add broad insight into the pathobiology of AAA due to the emergence of numerous transgenic and gene knockout mice.

Surgical Aortic Debanding: A Procedure to Study Left Ventricular Reverse Remodeling in Murine Model of Aortic Constriction

This video demonstrates the technique of aortic debanding, which can lead to reverse remodeling to limit the progression towards heart failure in a murine model. Post debanding, a significant reduction of ventricular afterload triggers regression of ventricular hypertrophy and fibrosis, causing a recovery in cardiac function.
