Adipose Tissue Quantification

Adipose tissue quantification is the measurement of body fat amount, distribution, and regional volume, providing clinically relevant information beyond body weight or body mass index. It commonly uses imaging methods such as computed tomography or magnetic resonance imaging to distinguish visceral from subcutaneous fat through tissue segmentation, while dual-energy X-ray absorptiometry and anthropometric measures provide complementary estimates of total adiposity. In medicine, these measurements support obesity assessment, metabolic and cardiovascular risk stratification, evaluation of body composition, and monitoring of responses to diet, exercise, medication, or surgery. Standardized quantification also strengthens research on how fat distribution contributes to disease.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.

This video demonstrates a technique for isolating and culturing fluorescently labeled neural crest-derived adipose-derived stem cells (ADSCs) from murine periaortic arch adipose tissue. Following isolation, the NCADSCs are detected, sorted using fluorescence-activated cell sorting, and subsequently cultured on a plate, promoting cell proliferation.

Isolation and Differentiation of Adipose-Derived Stem Cells from Porcine Subcutaneous Adipose Tissues

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

Videomicroscopy systems are used to examine functional properties of isolated adipose tissue arterioles in response to physiological and pharmacological stimuli. This technique can be used to examine microvascular phenotypes in different adipose tissue domains in obese humans.
