Athymic Mouse Model

Athymic mouse models are laboratory mice with an absent or severely underdeveloped thymus, making them deficient in mature T lymphocytes and valuable for studying human disease. Because impaired cellular immunity reduces rejection of transplanted human cells and tissues, researchers can implant tumors, organoids, or other xenografts and monitor their growth, treatment response, and biology in vivo. These models support cancer research, drug development, transplantation studies, and investigations of human tissue function, while their immune limitations require careful interpretation and, when needed, combination with complementary models that better represent the human immune system.

This article describes the generation of an orthotopic mouse model of human pleural mesothelioma by implantation of H2052/484 mesothelioma cells into the pleural cavity of immunocompromised athymic mice. The longitudinal monitoring of the development of intrapleural tumors was assessed by non-invasive multimodal [18F]-2-fluoro-2-deoxy-D-glucose positron emission tomography and computed tomography imaging.

Mouse Models of Periventricular Leukomalacia

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.

Mouse Models for Graft Arteriosclerosis

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.
